Howdy. In this video, Rob discusses how one can add NVMe addon cards to this NAS:

According to the Synology website, under the specs and accessories section, this device now has "M.2 SSD Storage Pool Support" but none of the NVMe add-on cards are supported, so you just have a choice of using the internal NVMe as cache, or storage pool, but not both.

Can you confirm DS1621xs+ no longer supports NVMe add-on cards?

Yes, you need to use internal M.2 slots for caching or storage (not both). You could use a couple of HDD bays for SATA SSD and use NVMe for caching. But in real life, NVMe storage is all you need. Caching simply boosts HDD RAID when SSD volume is not affordable.
